2023.03.21 03:00 LasagnaLeoGeck Helping scared babies and intimidating. I’m stuck
So, one of my males died from a sabertooth, and the babies went to hide in a little hide. I found exactly where they are, but there are predators near, so my ape is always in a state of panic, but not a dopamine drop, just the vigilant sign with the orange eye. I can’t get the babies out of the cave since my only option is to intimidate, since I’m on Xbox and it’s the same button. I’ve been trying for hours, and it seems to be stuck in a constant state of fear. That bongo music is getting on my nerves. I’m the first Evolution stage since it’s my first play through, and my apes are too dumb to make weapons. What do I do? I just want my baby apes, so I can continue the game

2023.03.21 03:00 SIIMythicWolf Shoot the Stasis Crystal Or The Frozen Enemy?
I've been playing for a few years now, and I am indeed a Titan main, I've recently decided to take a dive into stasis, but as far as I look, I seem to not be able to find an answer to a burning question, when I throw my glacier grenade and enemies get frozen near it, do I get more damage from shooting at the frozen enemy? Or by shooting the stasis Crystal and it bursting to the frozen enemy? And in boss fights, are stasis crystals worth spawning on the boss to damage them?

2023.03.21 02:59 TheUpsideDownBarnOwl I just got dumped, but I'm not exactly upset.
Tldr: I was in a relationship where I had to change who was I was to make her like me, and the two of us were near opposites.
About two months ago I got with this girl in liked way back when. Two years ago she suddenly cut me off and then gave the me silent treatment, without explanation. We ended up talking again roughly two months ago and started dating within two weeks.
She was my best friend, and helped me through some tough times. She understood my mental health and my mood swings, and even tolerated them. However, I treated her like shit. I reflected my own insecurities, like how I didnt trust her and how I felt her family didnt like me onto her. She broke up with me Thursday but I got her back yesterday. I promised to be fully open with her and change how I acted.
Not much more than a day later I got disrespectful to her (very disrespectful) friend after she got uppity with me while my blood sugar was through the floor and I was on the edge of unconsciousness. I directly broke the promise of becoming a better person within a DAY. I tried apologizing for it later but she just told me she couldn't be with me and didn't have feelings for me anymore. I knew there was no third chance.
At first I was hurt, as anyone would. I really did care for her, and felt terrible for it. However after a few hours past I suddenly didn't feel it anymore, and felt oddly happy.
I felt happier than I was when I was with her, than I ever really have. I learned some lessons, learned what I need to fix with myself and what not to do in the future, but I guess I wasn't as happy as I thought with her.
In hindsight, we were too different to really stay together very long. We were both blinded by our feelings for each other to realize each other's (and our own) faults and how they conflicted.
I sort of felt like I had to walk on eggshells around her and be a cuddly teddy bear which I really ain't. I feel like I would have to change everything about who I am to be with her.
I know I treated her wrong when I got caught behind my insecurities. I broke a promise the day after making it and proved I couldn't change, but I dont feel bad about losing her.
So here's tip for everyone falling for someone, take a step back. Think of both of their faults and how you'll have to adjust to them, and think of what kind of person you are. Otherwise, the both of you will be hurt. Also, dont fully change who you are to be with someone because that change will not stick.
Instead, your mind will. You'll get so absorbed into the feeling of being a relationship that you'll forget how you feel, and if you're unhappy. I was fairly happy with her, holding her, driving her home with one on the wheel and the other around her in the bench seat of my pickup. I enjoyed every little kiss, but I didnt enjoy having to change who I was.
Find someone who loves you for you. You'll both have to adjust how you act for the other, but dont change who you are to make someone love you. If you want to change who you are, change for you and you alone.
And the last bit, make sure you're ready for a relationship. Taking her on dates to town in an old truck that gets 11 miles per gallon, and cost $75 to fill up (in this economy), took a lot of my measly paycheck. Combine that with the fact I wasn't mentally prepared as I hadn't rebuilt my trust from the two year hiatus, I quickly went into feeling like she was an enemy.
Frankly, shouldn't have gotten with her to begin with. We weren't meant for each other, no matter how much we would have tried to make us work, our personalities conflicted and it was very easy to hurt her, even when trying not to. While I feel guilty about how I made her feel, I'm not so sad about being getting out of the relationship.
Lessons were learned and life rolls on. I'm going to work on myself as a person, but I ain't going to try to get her back. All we ever did was fall in and fall away, heartbreak for both of us.

2023.03.21 02:59 LHFE Stray Cat’s Tail Degloved
As noted, a stray kitten my family has been carrying for had his tail degloved. I bought some MicrocynAH 10 days ago — purchased the morning after the injury — because I don’t live nearby and stores were already closed when it happened.
The cat is skittish so we could never get to close except to spray the tail, but the baby let me near him today. His tail looks good on the surface, but when I separated the fur a bit I could see what appears to be a full inch of exposed bone that is turning brown. We didn’t know there was any bone or ligaments exposed until tonight because he’s warmed up to me enough to pet him and check. He’s been forced to live in the basement since the incident.
I don’t know what to do. I was laid off from work today and can’t afford an expensive surgery. He is the sweetest kitten I’ve ever known (and I’ve known and worked with a lot of cats as a foster), so I know he would fair well with a family — he’s just scared cuz he’s always lived outside but I’ve always paid for his food, and my mom always gives him attention.
I know this is an emergency, so my options are either have him put down because I can’t afford his surgery, give him to a local shelter that will still put him down, or find a service that will help pay costs.
I want to find a low-cost surgery center anywhere in Virginia/DC/WV/MD area. I’ll drive as far as I need to. I don’t want this sweet baby to die, but I don’t want him to suffer either.
Please help Me! I know he needs a vet, so please don’t say “VET NOW” because I already know. He’s going somewhere in the morning, I just don’t want that visit to end in his euthanization.
Thank you for any help!

2023.03.21 02:59 LunarIslands Angry at my husband’s surgeon
At the gentle recommendation of my husband and some kind Redditors, I am in the process of trying to find a professional to help me through this. But I just have to get this off my chest.
Recently, my husband had surgery to repair his ACL and MCL. Before we left for the hospital, he took a COVID test and checked his temperature as we were told. The COVID test was negative, but his temperature was 99. I know that’s not alarmingly high for anyone, but for him this is nearing a low-grade fever because his baseline is in the upper 96’s, low 97’s.
We reported this to the surgeon. She said she was not concerned because my husband felt well overall. She did draw some blood and said she didn’t see any signs of infection.
The surgery itself was successful. At home, my husband took a nap. His pain was managed and he was feeling fine, just groggy and quiet. I checked in on him an hour later to find him sweating bullets and shivering in his sleep. I woke him up and checked his temperature. It was now up to 103, which for him is miserably burning. He was also saying his back was hurting him terribly.
We went to the ER and he received IV antibiotics and oxygen. Once he was feeling better he was discharged home once more.
It seemed to not bother him too much, though I know he processes things emotionally at his own pace. For the next week I was an emotional mess. Prior to this I’ve seen my husband have the occasional headache and sniffles. He’s never sick sick. So this was especially jarring.
At night I would wake up thinking I was hearing him shivering again. At random intervals during the day I would get teary-eyed and leave to cry for a few minutes. I tried to keep this from my husband because I didn’t want him to worry while he was recovering. He eventually caught on.
I felt somewhat better after finally crying into his shoulder and just having him hold me for a while. I was still emotional from time-to-time but was managing better.
We met with the surgeon today to make sure my husband’s knee is healing well so he can start physical therapy. It is. I’m not. My face was literally red for the entire appointment. I don’t doubt that the doctor is a good, caring lady. She was very attentive post-surgery. But I feel so much anger and I blame her for causing my husband so much pain by brushing off what we told her was a fever.
Reddit was very helpful in reassuring me during the sadness. I guess I am looking for reassurance now during the anger. I just don’t know what to do other than rant it out.

2023.03.21 02:58 RogueGuardianStudios Just got back from 7 days on the Carnival Celebration
Just got back off of Carnival newest ship the Celebration, and boy oh boy is it a mixed bag.
First things first, there were 4 of us in a family harbor suite me and my wife (mid 30's) and our two kids (7 M, and 22mo F).
The room was adequate and had plenty of storage. The beds were comfy in the hotel kind of way. The only issue with the layout was with the door to the room being making it very hard to get our stroller in and out on port days.
The cleanliness of the room left much to be desired, as my little one found leftover peanuts from the previous guests on the first night in the room. These were just loose on the ground not in a package or anything like that. As time went by I also found where some soda had splashed one of the cabinets and had dried into a syrupy mess, also there before we got the room. The bathroom doors handle already had several screws loose and was coming away from the door.
The bathroom itself was cramped, but the shower had plenty of room and I never felt crowded will showering (I'm a larger guy 5'9 230 lb), but that did come at the expense of the rest of the bathroom which felt very crowded.
My son enjoyed the water slides on the back of the ship, except for one which was always out of service (the drop slide), and while he was not tall enough to ride the Bolt roller coaster it wouldn't have mattered since it was down for maintenance most of the time. However he ended up spending most of his time in Camp Ocean where he had a blast.
my daughter on the other hand enjoyed her time, but was not as well taken care of. Celebration offers "Turtles" level of care which is 0 - 1. This was a big selling point for us as we have cruised with Royal and they have nursery care as well. The hours for this care however where horrible. I tried to look up the hours before the cruise but they were not posted anywhere. On sea days they would watch her from 8am - 10am (maybe), and 10pm - 1am, and on port days it was 3-4 hours of the port stay but off set in away that you would miss about half of the day if you used it (we did not), then the 10pm - 1am all of this at $9 hr + tip. Considering how much of a selling point this was and how much Carnival was hyping it up this is disgraceful. While I'm not saying we should be able to drop her off all day, the availability made it impossible for me and the wife to have a nice dinner or catch one of the shows just the two of us. This was not an issue on Royal.
The food was a mixed bag. The main Dining room never had anything that made me go wow, except for their melting cake desert. However Guys burger joint was wonderful greasy goodness, Big chicken and some of the best chicken I have had in a long time, and The Pig and Anchor was amazing as well. The steakhouse was not good or worth the extra charge. We skipped the seafood restaurant. The "pop up" street eats where always good, and I love the idea of a new small themed snack everyday. I ordered the Seafood shacks buffalo shrimp one time, and while it was made to order and very good, it was not buffalo. It was more of a thai chili sauce. The pizza was good late night food and always a kid pleaser. Deco Deli's sandwiches where all good as well. The sushi Restaurant was hit or miss, but that is not an thing I normally do, and my wife loved everything there so take that for what is worth. The Japanese steak house was wonderful, large portions of good food with a light hearted show to go with it. However the best food hands down was at Emeril's. Everything was amazing, not a bad thing on the menu.
The Casino was there. I am a big Crapps player, I enjoy the atmosphere and getting to know the other players. Carnival did not make this easy. First off it was a $15 table (other lines are $10, they said it was a big ship tax...), second only about a third of the dealers knew what was going on( this caused 10 plus min waits between rolls sometimes ) and they would constantly misplace your bets or not pay you out right. This caused issues, and regularly spoiled the atmosphere at the table. Third pretty much anything goes with the dice. Throw it just over the halfway point on the tabe no where near the back wall, no issues. Toss the dice over everyone's heads, no issues. Hit people with the dice! As long as the dice landed back on the table, it was all good. At one point in time someone threw in their money and card as the shooter was throwing and got hit by the dice and it was counted. I stayed because it was the only game in town, but never again.
And we have to talk about the kids. At all hours of the day and night there were packs of kids roaming the Lido deck, buffets, and other halls. Most of the time this was not an issue, however when it was it was because a 15 -17 yr old where fighting in the buffet (Carnival did not break it up, and this "play" fighting bumped into several other guest as well as knocking over a small child) or a group of 10-12 yr olds running through the halls bumping into other guests screaming profanities the whole time. I don't know the solution to this but I will not be coming back so I don't have to worry about it.
Overall the staff were always kind, except for one person who gave me an "that's not my job" when asking for replacement silverware. Apparently their shift had ended, but they were still in full uniform.

Have been flying this route for work the past few months and several flights are on 787’s with business class (though the service is nowhere near Delta One or Polaris). Is anyone familiar with this route and if this is just a seasonal thing? Looks like it stops next week, but I can’t find anything about it.

2023.03.21 02:56 toughToFindUsername Kayak recommendation
Hi there, long time lurker and owner of a small 9ft Islander sit on top kayak. After over a year on the waiting list, i finally got a dinghy space in the marina near my home. I want to get something bigger, at least 11ft. But I'm unsure of what brand or sit on top vs in. I'm a big guy and plan on riding in the marina and the ocean as well as the river that connects. Any advice on specs and brands would be appreciated. I'm thinking spending around $1,200 but curious what y'all think a great option would look like.
Edit: I'm located in California

2023.03.21 02:56 Sea-Opportunity7595 Tips for line person
Been at chipotle about a month now and still feel pretty stupid on line. Although, I have by myself gone through a whole line going outside the door during peak. My biggest struggles are wrapping burritos fast and neatly, keeping line nearly completely neat while putting everything on bowl/burrito, and just how to get faster (already do things like ask people for things they want while tortilla is still in press or while grabbing bowl.) Any help would be great thanks!

2023.03.21 02:56 elcapitan58 My friend cut me off out of nowhere and I’m completely heartbroken.
On mobile so apologies for formatting.
Title pretty much says it all.
I was having a rough week last week and a very good friend of mine invited me to visit her for a bit. It was a fun time, but some stuff happened near the end, and I ended up leaving in a very emotional state due to a panic attack without saying bye.
I got home to discover myself blocked on the app we primarily use to communicate. For whatever reason, I wasn’t blocked on a different one, but all that’s really doing is giving me false hope I can fix things later on.
I have no idea what truly happened and I’ve been racking my brain for the past 48 hours to no avail. I have so many theories and any of them could be true. The worst part is that there’s legitimately nothing I did that I believe warrants being cut off without explanation. It’s just stunning me. And it came literal hours after she talked about how much she trusted me.
I’m just heartbroken. Truly devastated. We didn’t even talk. I just apologized over text and asked if we could discuss what happened the next day. Never happened. Got this instead and it’s tearing me apart.
I’m not used to this feeling either. I’ve only lost 2 friends in the last 3 years, and I wasn’t this close with them. What a horrible feeling this is.
Life just sucks sometimes. Some people are telling me to give them some space and try to contact again in a week or two. But it might be over. My heart is legitimately in pieces.

2023.03.21 02:56 piroskamcs Corporatism of Vets
This was in our local news today (NZ)
Overseas private equity companies are buying up vet practices in New Zealand and branding them as ‘local’. Independent Kiwi vets are fighting back
Throw “your local pet vet” onto Google and you’ll get a few options. A vet clinic in Hillcrest, Hamilton, which “can boast its origins back as far as the 1960s”. A Manurewa, Auckland practice “serving the community for over 40 years”; Taradale Vet Hospital “established in Napier” 40-plus years, The Gardens Veterinary Clinic practising in Dunedin for over 30 years.
But what their pet owner customers may not know is these vet practices, plus a couple of hundred others in New Zealand and Australia, all trace their ownership to Germany.
That’s where the Reimann family, one of the wealthiest families in the world, comes from. Four family members head a vast (more than US$50 billion) private equity network headquartered (almost certainly for tax reasons) in neighbouring Luxembourg.
The Reimann investment company is called JAB. It owns a variety of consumer brands – Krispy Kreme doughnuts, Dr Pepper drinks, Bally shoes, Coty cosmetics, the British sandwich shop chain Pret a Manger, and Espresso House, the largest coffeehouse business in the Nordic countries.
It also owns pet businesses - insurance companies and vets.
NVA owns VetPartners, the biggest vet company in Australia, and through that holding, JAB is the proprietor of about 50 New Zealand vet practices, including the Pet Doctors vets.
And the vet business is seen as relatively recession-proof.
'Should you trust private equity to take care of your dog?’
Yes and no, says Dr Megan Alderson, who co-owns and runs The Strand Vet in Parnell, Auckland.
Historically, corporates have had a bad reputation with some vets and owners alike. Independents have been portrayed as more patient-centric, with corporates portrayed as placing more emphasis on financial success.
In the vast majority of cases, these corporations take measures to limit the authority of the practitioner, which often gets in the way of their ability to administer quality care. In fact, it’s relatively common for parent companies to assign mandates that are meant to be carried out universally, regardless of an animal’s individual needs or best interests.
John Robb, an exemplary veterinarian went into business with Banfield because of the financial security it would provide for his family.
Unfortunately, Robb’s prosperity didn’t last for long. Now entangled in a bitter legal battle with Banfield, Robb claims that the corporation stole his hospital and “is turning animal medicine into an exploitative, even dangerous business.” According to Robb, Banfield and other businesses like it are committing ‘peticide,’ or “the systematic destruction of pets by corporations for profit.”
In 2012, Banfield claimed that Robb violated its vaccine protocols and then seized his practice. Robb’s defense? It’s true that he was giving some of his smaller patients half-doses of vaccines, but he did so after several of them had nearly died from vaccine-related reactions.
Despite the corporation’s claims that Robb was operating dangerously, his practice’s mortality rate is significantly lower than many of the vets Banfield employs.
According to 2015 data, for example, Robb had a 100% survival rate in 2,241 canine dental procedures, whereas Banfield lost 747 dogs in five years, “about one death for every 2,000 dental procedures.”
Banfield operates unnecessarily, often sedating dogs every year for an “annual cleaning,” which is an entirely elective (and often superfluous) procedure.
And other vet Bragdon is skeptical. She says it can be tough having realistic conversations about what operations are and are not worth having on their pets.
Bragdon recalls an owner bringing in a dog with heartworm disease that had a very low chance of survival. The owner wasn’t sure what to do. Bragdon tried to put the cost and risk in perspective.
“I told the client, if you can’t afford to go to Las Vegas and lose $500 to $1,000 gambling, and that $500 to $1,000 means you can’t pay rent, you can’t pay your car bill, you can’t feed your kids, we need to humanely euthanize this dog.”
Conversations like these, she says, are not be the norm at a corporate clinic.
I think I agree. We changed vets a while ago with our previous cat. The vets seemed uninterested in doing follow up checks and more interested in pushing the "special" food. New vet, an independent, was comprehensive. Not only getting the notes from previous vet, but spending quite some time, explaining things in detail to me on the phone! He gave me a link to a vet website, where you can look things up (and not get the dubious content), and prescribed some new medication for cat. After that she only needed some supplementary probiotics added to her food, not "special: food either.
I wonder if corporate vets push the things involving money more.
Another vet I used to go to, he had a small clinic, no flash expensive equipment and such, no hordes of helpers, and was the best surgeon I had ever come across. Amazing diagnostics too. And he had the hard talk with me with one of my cats. Sadly I took her to another, who confirmed his view and I had to have her put down. Should have listened shouldn't I< but still it makes me wonder.
Corporate vet wanted to do things, and why? It wasn't a curable issue.

2023.03.21 02:55 Mission_Mix3665 I just don't have tolerance for being yelled at
I really just don't. I do not feel any vengeful feelings, all I feel is I want nothing to do with you from here on out. One time is all it takes for me to want to sever any future contact with the person. It's super petty but really all it takes is one time for me to want to never have to talk to you ever again.
I don't harbor any negative feelings. Please go on and win the lottery, marry the most amazing person, have your life be the most incredible thing ever but I just never want to interact with you ever again. You have become in my eyes a grenade that can go off at any point. You have taken yourself out of the group of people I feel safe or comfortable around and put yourself into the threatening group so that I just do not want to interact with you ever again.
That's the thing, zero negative feelings for the person, just that I have now come to view you as a threat and once that happens, I don't want you anywhere near me. I will go a long way to cut someone out of my life when they've crossed that line, literally so far as moving to avoid the person it's that strong the feeling of them being someone I never want to interact with again.
The biggest reason for that is people who go around yelling at others seldom ever realize how horrible they are, you can say to them, dude your yelling at people is super messed up and they will just come up with tons of reasons why you deserve to be yelled at, why they are in the right and probably just get angrier and yell at you more. So that there is so resolution to that person except removing yourself from any interaction with them and with time forgetting they exist.
I say all this as someone who myself has a temper that I continually must battle but realizes that yelling at people is the very last thing you ever should do. If I get angry, I vent in designated venting areas online, go to a place no one is around and curse, run until I vomit, basically I know how fucked up losing my temper is and so don't fucken do that shit. What I've realized is the people who go around yelling at people do not realize what they are doing is fucked up. They are convinced they are in the right and once someone is convinced of that, you cannot do anything for that grenade but throw it away from you and let it destroy itself.

2023.03.21 02:54 DepressedAndObese Sudden poor performance - help me diagnose it?
I have been working on a scene for about a year in my spare time, it's quite heavy (multi millions tris), and I'm on the best gaming laptop I could afford a fair few years ago so I'm not expecting blistering performance, however lately it has been verging on unusable.
Started in January, I updated my Geforce driver from 527.56 to the latest one and it's just slow. The render view viewport doesn't load in properly, and sort of stops after 3 samples, never cleans up any further. Renders freeze while building the bvh and crash a lot of the time. It just generally seems to be chugging.
I wondered if it's a hardware issue, as despite it being a heavy scene, it was all ok before, lots of instancing, simplification anyway, and I even went in and optimised everything and shaved nearly a quarter of the tris with remodelling and decimate modifiers.
I'm fearing the hard drive might be going, does that make any sense to anyone? Like it's having difficulty pulling the textures etc because the HDD is bad? I'm not getting any warnings in Windows but CHKDSK sfc/scannow etc is all I've done. If not the HDD maybe the GPU? I did have a couple of BSODs around the update but I went back to the previous Nvidia driver and then waited for a later update in Geforce Experience before I tried updating again, and since that update system stability is fine, but Blender is still slow.

2023.03.21 02:53 anarchodelphis Crunfli et al, 2022: Gray matter loss after "mild" COVID-19 was associated with ↑anxiety, ↓verbal memory; SARS-CoV-2 found to be able to infect cultured astrocytes; infected astrocytes seem to produce secretions that 'kill' uninfected, cultured neurons.
| https://www.pnas.org/doi/full/10.1073/pnas.2200960119 TL;DR in last paragraph. “In line with the potential neurotropic properties of SARS-CoV-2, recent evidence indicated the presence of viral proteins in brain regions of COVID-19 patients ( 21, 22) as well as in the brains of K18-ACE2 transgenic mice ( 22, 23) and Syrian hamsters ( 24) infected with SARS-CoV-2. The presence of SARS-CoV-2 in the human brain has been associated with marked astrogliosis, microgliosis, and immune cell accumulation ( 21). Further indicating the ability of SARS-CoV-2 to infect cells of the CNS, SARS-CoV-2 has also been shown to infect human brain organoid cells in culture ( 22, 25– 27). Recently, SARS-CoV-2 has been found to cross the blood–brain barrier (BBB) in mice ( 28– 30) and in two-dimensional static and three-dimensional microfluidic in vitro models ( 31, 32), therefore potentially reducing the integrity of the BBB. We performed a cortical surface–based morphometry analysis (using a high-resolution 3T MRI) on 81 subjects diagnosed with mild COVID-19 infection (62 self-reported anosmia or dysgeusia) who did not require oxygen support. The analysis was performed within an average (SD) interval of 57 (26) d after SARS-CoV-2 detection by qRT-PCR, and the subjects were compared with 81 healthy volunteers (without neuropsychiatric comorbidities) scanned during the COVID-19 pandemic (balanced for age [ P = 0.97] and sex [ P = 0.3]). The COVID-19 group presented higher levels of anxiety and depression symptoms, fatigue, and excessive daytime sleepiness ( SI Appendix, Table S1 shows epidemiological and clinical data). An analysis of cortical thickness (adjusted for multiple comparisons using the Holm–Bonferroni method) revealed areas of reduced cortical thickness exclusively in the left hemisphere, including the left gyrus rectus (P = 0.01), superior temporal gyrus (P = 0.036), inferior temporal sulcus (P = 0.02), and posterior transverse collateral sulcus (P = 0.003) (Fig. 1A). There was no observed increase in cortical thickness. A subgroup of 61 participants of the COVID-19 group also underwent neuropsychological evaluation, which assessed episodic verbal memory (logical memory subtest, immediate and delayed recall, Wechsler Memory Scale), sustained attention (Color Trails A), and alternating attention and cognitive flexibility (Color Trails B). The tests were performed a median of 59 d (range between 21 and 120 d) after diagnosis; we observed fatigue in ∼70% of individuals and daytime sleepiness in 36%. Despite the high level of education of the participant subgroup (median of 16 y), the comparisons with Brazilian normative data ( z scores were adjusted for age, sex, and education) showed that nearly 28% of participants presented impairments (z scores ≤ −1) in immediate episodic verbal memory (immediate recall, including mild, moderate, and severe impairments), and ∼34 and 56% underperformed (z scores ≤ −1) on Color Trails A (sustained attention) and B (alternating attention and cognitive flexibility), respectively (SI Appendix, Fig. S1 and Table S2). Interestingly, 77% of these COVID-19 patients also presented acute anosmia or dysgeusia, which may be related to the observed changes in cortical thickness ( 19). The high proportions of anosmia and dysgeusia support the idea of the virus entering the nervous system, more specifically the orbitofrontal region (due to the proximity and communication with the nasal cavity). We observed a negative correlation between BAI (Beck Anxiety Inventory) scores and the cortical thickness of orbitofrontal regions (adjusted for fatigue scores) ( Fig. 1B and SI Appendix, Table S3). We also identified a positive correlation between immediate episodic verbal memory and cortical thickness of regions associated with language (adjusted for fatigue scores) ( Fig. 1C and SI Appendix, Table S3). There was a likely correlation between Color Trail B and the thickness of the gyrus rectus after adjusting for fatigue ( r = 0.2, P = 0.13). Our findings indicate that cortical thickness atrophy is associated with neuropsychiatric symptoms and cognitive impairment in COVID-19 patients with mild or no respiratory symptoms. Figure 1: Cortical thickness atrophy after mild COVID-19 infection. Surface-based morphometry by high-resolution 3T MRI (A). Results from the analysis of 81 subjects with confirmed SARS-CoV-2 diagnosis (who had mild respiratory symptoms and did not require hospitalization or oxygen support) compared with 81 healthy volunteers (without a diagnosis of COVID-19). The analysis was performed within an average (SD) of 57.23 (25.91) d after diagnosis. (B) Correlation between anxiety scores (BAI) and right orbital gyrus thickness. (C) Correlation between Color Trail B test (Z-TRAILB: z-scores were based on Brazilian normative data) and left gyrus rectus thickness. Data depict partial correlation coefficients (adjusted for fatigue). [...] To investigate proteome-level changes caused by SARS-CoV-2 infection, we conducted a liquid chromatography–mass spectrometry (LC/MS) proteomic analysis with a separate set of samples consisting of 12 postmortem brain samples from COVID-19 patients vs. 8 SARS-CoV-2–negative controls ( SI Appendix, Tables S6 and S7). We identified 656 differentially expressed proteins: 117 down-regulated and 539 up-regulated. Pathways associated with neurodegenerative diseases, carbon metabolism, and oxidative phosphorylation were enriched among the differentially expressed proteins (SI Appendix, Fig. S6). Notably, proteins expressed in astrocytes were enriched among the differentially expressed proteins, consistent with the higher frequency of infected astrocytes observed in COVID-19 postmortem brains ( Fig. 2F). To gather further evidence of the susceptibility of human astrocytes to SARS-CoV-2 infection, neural stem cell–derived human astrocytes (BR-1 cell line) ( 35) were exposed to the virus, and the viral load was determined at 24 hpi ( Fig. 3A). Confirming what was seen in the postmortem brain samples, viral genetic material (Fig. 3B), dsRNA, and spike protein (Fig. 3 C and D) were detected in infected cells with significant overlap (Fig. 3 C and E). SARS-CoV-2 infection reduces cell viability by over 25% at 72 hpi (Fig. 3F). To confirm the ability of SARS-CoV-2 to infect human astrocytes, we used a replication-competent eGFP reporter–pseudotyped vesicular stomatitis virus (VSV) in which the glycoprotein gene (G) of VSV was replaced by the full-length SARS-CoV-2 spike (S; VSV-eGFP-SARS-CoV-2) ( 36). The human astrocyte culture was also infected by VSV-eGFP-SARS-CoV-2 (Fig. 3 G and H). Altogether, these results provide strong evidence that human astrocytes are permissive to SARS-CoV-2 infection and also represent a site for viral replication in the CNS. [...] …we started by using a publicly available, single-nucleus RNA-sequencing (snRNA-seq) dataset from brain samples of patients with COVID-19 ( 37) to analyze the mRNA expression of the classical SARS-CoV-2 receptor ACE2 as well as the alternative receptors NRP1 and BSG ( 38– 40). These analyses revealed that ACE2 mRNA was undetected in astrocytes ( 21, 37); however, astrocytes did express detectable levels of NRP1 and BSG mRNA (Fig. 4 A and B). We also found that the levels of expression and percentage of astrocytes expressing NRP1 and BSG mRNA were increased in astrocytes from COVID-19 patients compared with controls ( Fig. 4 A and B). [...] …we performed western blotting using cultured neural stem cell–derived astrocyte extracts to evaluate if the data from snRNA-seq matched proteome-level data from our in vitro model with ACE2-transduced A549 cells as the positive control. The human astrocytes used in this study did not express ACE2, whereas they did express NRP1 ( Fig. 4 C–E and SI Appendix, Fig. S7). To determine whether NRP1 was the receptor permitting SARS-CoV-2 infection in astrocytes, we preincubated these cells with a neutralizing anti-NRP1 antibody. Neutralization of NRP1 inhibited the infection of cultured astrocytes by SARS-CoV-2 ( Fig. 4D) as well as the infection of cultured astrocytes by VSV-eGFP-SARS-CoV-2 ( Fig. 4 E and F). These results confirm that SARS-CoV-2 infects human astrocytes via the NRP1 receptor. [...] …we analyzed the proteome of SARS-CoV-2–infected human astrocytes. LC/MS-based shotgun proteomics revealed 170 differentially expressed proteins in SARS-CoV-2–infected astrocytes compared with mock controls… [...] …pathway enrichment and interactome analyses predicted a wide range of biological processes and regulatory networks affected by SARS-CoV-2 infection ( Fig. 5A). Pathways involved in carbon/glucose metabolism were among the most enriched ( Fig. 5B). More specifically, proteins found differentially expressed in SARS-CoV-2–infected astrocytes and in COVID-19 postmortem brain tissue samples belonged to glycolysis/gluconeogenesis, carbon metabolism, and the pentose phosphate pathway (Fig. 5C). Collectively, these data reinforce that SARS-CoV-2 infects astrocytes in the CNS, possibly affecting energy metabolism pathways and modulating proteins associated with neurodegeneration. [...] Astrocytes are essential to brain homeostasis not only because they are the main energy reservoirs of the brain ( 42) but also, due to their important role in protective responses to cell damage triggered by infection or sterile inflammation ( 43, 44). There is evidence that astrocytes may secrete yet undetermined neurotoxic factors ( 43– 45) and are also involved in the uptake, synthesis, and distribution of brain metabolites ( 46, 47). Thus, we investigated if neuronal viability could be affected by exposure to media conditioned by SARS-CoV-2–infected astrocytes. To test this hypothesis, we cultured NSC-derived neurons or differentiated SH-SY5Y neurons for 24 h in a control or conditioned medium in which SARS-CoV-2–infected astrocytes were grown ( Fig. 6A). The conditioned medium increased the rates of apoptosis by 45.5 and 22.7% in NSC-derived neurons and SH-SY5Y neurons, respectively ( Fig. 6 B–E and SI Appendix, Fig. S10 A and B). The possibility of neuronal infection was ruled out as viral RNA was not detected in either cell type after exposure to the conditioned medium ( SI Appendix, Fig. S10 E and F), and direct exposure to SARS-CoV-2 did not reduce the viability of NSC-derived neurons or SH-SY5Y neurons after 24 ( Fig. 6 D and E), 48, or 72 h ( SI Appendix, Fig. S10 C and D). These results suggest that SARS-CoV-2–infected astrocytes release soluble factors, which reduce neuronal viability. Figure 6 (cropped): Percentage of living or nonliving (D) NSC-derived neurons and (E) SH-SY5Y cells. Cells were classified as living (gray bars; double negative), in early apoptosis (purple bars; apotracker+/FVS−), in late apoptosis (pink bars; double positive), or necrotic (green bars; apotracker−/FVS+). (F) Differentiated SH-SY5Y cells were cultured for 24 h in the presence of medium conditioned by SARS-CoV-2–infected astrocytes (ACM CoV-2) or uninfected astrocytes (ACM CTL). The viability of SH-SY5Y cells was assessed using an ATP (adenosine triphosphate) -quantifying, luminescence-based cell viability assay (CellTiter-Glo) at 24 h postinfection. P values were determined by one-way ANOVA followed by Tukey’s post hoc test. **P < 0.01 compared with the mock group; ***P < 0.001 compared with the mock group; ****P < 0.0001 compared with the mock group [...] ...we found that SARS-CoV-2 infection elicits a neurotoxic secretory phenotype in astrocytes that results in increased neuronal death. A similar phenomenon has been observed when astrocytes are activated by inflammatory factors ( 43, 86, 87). The alterations in cortical thickness we observed in COVID-19 patients could be explained by this neuronal death, at least partially, as well as by other mechanisms, including reactive astrogliosis and alterations in astrocyte specification and morphogenesis, as previously described in Alzheimer's disease, autism, and schizophrenia ( 88– 90), as well as epilepsy, autism, and self-injury ( 91– 94). Matschke et al. ( 21) reported astrogliosis in 86% of individuals who died following a diagnosis of SARS-CoV-2 infection. In agreement, snRNA-seq data show that the main markers of reactive astrocytes ( 95) are enriched in samples from the medial frontal cortex of patients with COVID-19 compared with noninfected patients, supporting the hypothesis that reactive astrogliosis is a feature of COVID-19 ( SI Appendix, Fig. S11). Astrocytes are also relevant in the regulation of synapses (and neural networks) and have been linked to the manifestation of depression ( 96), anxiety symptoms ( 97), and memory impairment ( 98), all of which have been observed in our post-COVID infection cohort. In summary, our findings are consistent with a model in which SARS-CoV-2 is able to reach the CNS of COVID-19 patients, infect astrocytes via NRP1 interaction, and secondarily impair neuronal function and viability. These changes may contribute to the alterations of brain structure observed here and elsewhere, thereby resulting in the neurocognitive and neuropsychiatric dysfunctions manifested by some patients with COVID-19. Our study comes as a cautionary note that mechanisms of neuroinvasion in fatal COVID-19 could also be operative in mild COVID-19. However, it is important to note that the study was limited in that neuroimaging and cognitive testing were obtained from a different cohort than fatal COVID-19, in which only a minority of individuals showed evidence of astrocytic invasion.” submitted by anarchodelphis to COVIDZero [link] [comments] |
2023.03.21 02:53 Justice_Cooperative I was recently baptized in the World Mission of Society Church of God. But I want to leave immediately after for what I've discovered.
Hello dear friends, I was just invited in the World Mission Society Church of God because I expected that it would only be a teaching about the bible, then I got a good time learning about the bible with their local church leaderecruiter, after he done teaching about the bible, he told me directly it that I needed an immediate baptism, I wasn't expected that and I only came for the teaching about their bible because I'm kind of interested at first. They really force me to do immediate baptism to save my soul, and it seems that they wouldn't allow me to go home if I'm not baptized, so I followed them, I signed the baptism form paper, write my name, my phone number and my address to get baptized, and then went home. But after that, I found some information that they were cult-like religious organization, they were forcing their members to cut ties with the family, doing an opressive tithes, sex trafficking and forcing their members to live in the shared housing or communes. This made me scared and immediately want to leave My questions is
- Is World Mission Society Church of God really a cult?
- I'm scared that if I'm going to leave they will going to find and kill me since their church and my house are just very near.
- I signed up, put my name, my number and address in their baptism form paper, would they use that against me?
- My family don't know that I was baptized by this church, that's why I want to leave this church immediately, would they allow me to do so?
- They even told me that if I'm dissapointed in their belief, I'm free to leave, however I cannot really trust them. My fear is that whether they will find me then kill me, blackmail or Kidnap me
Can I leave this Church immediately safely?

2023.03.21 02:53 WeekendDoWutEvUwant 2 questions I can't find answers for by search function
[Using PF right now]
1) What would you do if you only have one setup ready for a FC, but some jars are colonizing much slower? (None have stalled but some seem to be moving SO SLOW)
2) In the other FC that I do have ready, it's already full of GT & LGT cakes -- the GT has already flushed once, but for some reason the LGT only just started pinning -- and only a couple of them. Is it just genetics? Are the growing conditions that different? I don't see how they could be (but I will admit I didn't mist nearly enough the first 2 days in FC)
Otherwise, what might I have done wrong? The 2 sets of spores colonized roughly at the same time, within a month, and both sets of myc looked solid

2023.03.21 02:53 Jakejake100 Going to my first ever United game!
After being a fan from the United States for 14 years, I will be traveling to see the last game of the year in May. I am beyond excited, but I am super unfamiliar with the area. I was just wondering if there are any solid places to go before or after the game? Or any nice places to eat for dinner near the stadium? Thanks! Cannot wait to see everything in person.
